A lack of food, water, or power will bring down their mood. Assign them to the correct job. Dwellers are happier in rooms which correspond to their highest SPECIAL stat. A successful rush raises happiness for everyone assigned to that room by 10%, but failing one does the exact opposite.

You may also want to know how your dwellers are feeling: Their happiness contributes to the overall health of your post-apocalyptic sanctuary, and unhappy dwellers mean an overall decrease in your vault’s production. When it comes to your dweller’s health, you want to try and keep it at full whenever possible.
A good tip is to listen the people. If you zoom in the rooms, you will see them talking about stuff that bothers them or stuff they would prefer to do! If the dwellers have red on their health bar, they have radiation poisoning. Give them a radaway, and they will be happy again. Job Satisfaction.
What’s the best way to keep the dwellers happy?
Drag in some weapon-toting Dwellers and don’t let those problems fester. Get rid of corpses. Sometimes you’ll have Dwellers die. It happens. That doesn’t mean you have to let the dead bodies lie around. You’ve got two choices; either pay some Caps to restore the dead Dweller to life or chalk them up as an acceptable loss and clear them out.
How does a radio station make dwellers happy?
If you zoom in on a particular Dweller, they will talk, and say why they are unhappy. One of the tips in the loading screens said a Radio Station makes Dwellers happy. Not sure this increases the general happiness or only counts for those Dwellers assigned to the Radio Station. There might be other structures which will increase happiness.
